Maximum mTOR Igniting BCAA & EAA Matrix

Amino acids are organic compounds that combine to form proteins.  They are commonly referred to as the building blocks of protein.  There are 3 classes of amino acids; non-essential, essential and conditional.   Non-essential amino acids are naturally produced in the body whereas essential amino acids are not synthesized by the body and must come from diet and/or supplementation.  There are 9 essential amino acids (EAAs) which promote muscle protein synthesis (MPS).  Of course the Maximum mTOR Igniting BCAA & EAA Matrix includes all 9 EAAs plus l-tyrosine.  L- tyrosine has been found to have an acute effect in preventing a decline in cognitive function in response to physical stress.  

Among the nine EAAs there are three aminos known as branched chain amino acids (BCAAs); leucine, isoleucine and valine.  BCAAs have the greatest impact on MPS and fatigue reduction.  Skeletal muscle is a major protein source for catabolism and BCAAs are metabolized during this process so it is important that you are getting an adequate amount in your diet.  For this reason the Crazy 10 Anabolic Amino Blend contains a proportional amount of BCAAs with 5g leucine, 1.5g isoleucine and 1.5g valine.

A clinical trial was conducted to determine the effect of EAA supplementation with a higher amount of leucine content on postexercise skeletal muscle metabolism and MPS.  Study participants consumed an EAA drink with equivalent amounts of dietary nitrogen (10g) during exercise.  However, one group consumed an EAA drink with 3.5g leucine content (L-EAA) while the other group consumed an EAA drink with 1.87g leucine content (EAA). leucine.  MPS was 33% greater (P < 0.05) after consumption of L-EAA (0.08 ± 0.01%/h) than after consumption of EAA (0.06 ± 0.01%/h). Whole-body protein breakdown and synthesis were lower (P < 0.05) and oxidation was greater (P < 0.05) after consumption of L-EAA than after consumption of EAA.  These findings indicate that consuming an EAA supplement with a higher leucine content during endurance exercise elicits a greater MPS response during recovery.  ENDUR3 goes even further than the studied dose of 3.5g with a massive dose of 5g of leucine to maximize MPS.

As the following study demonstrates, supplementation with EAAs in the absence of regular exercise is still beneficial.  14 elderly women (68 +/- 2 yr) participated in a randomized, double-blinded, placebo-controlled study to assess EAA supplementation on postabsorptive muscle protein fractional synthesis rate (FSR), lean body mass (LBM), and one-repetition maximum muscle strength, and androgen receptor and IGF-I muscle protein expression.  The supplemented group (SUP) consumed 15g EAA/d and the control group ingested a placebo for a period of 3 months.  Ingestion of 7.5 g EAA acutely stimulated FSR in both groups at 0 months (P < 0.05). Basal FSR at 3 months was increased in SUP only. The magnitude of the acute response to EAA was unaltered after 3 months in SUP. LBM increased in SUP only (P < 0.05). One-repetition maximum strength remained unchanged in both groups. Basal IGF-I protein expression increased in SUP after 3 months (P = 0.05), with no changes in androgen receptor or total and phosphorylated Akt, mTOR, S6 kinase, and 4E-binding protein.  This study shows that supplementation with EAA has an acute anabolic response and is maintained over time and can improve LBM.

Fatigue Destroying Matrix

It is natural to feel sore and fatigued when you are pushing yourself to the limits and gaining muscle.  DemiGods have no tolerance for fatigue, we must ENDUR3 so we can go harder during the next training session.  Say goodbye to muscle soreness with the Fatigue Destroying Matrix.  It includes 2.75g of L-glutamine and 1g of taurine to minimize muscle soreness and fatigue.

Taurine is an amino acid that has a sulfonic acid chain attached to it, in contrast to other amino acids that have a carboxyl acid chain.  It is classified as an conditional essential amino acid, meaning it is essential only during periods of diseases or physical stress such as strenuous exercise.  However, your body not produce enough taurine on its own so it is important to obtain the amino sulfonic acid through diet and/or supplementation. Taurine has several benefits including restoring insulin sensitivity, mitigating diabetic complications, reversing cardiovascular disease factors, preventing and treating fatty liver disease.  Most pertinent for athletes is its ability to reduce muscle soreness and cramping.

Thirty-six untrained male subjects (22.5 ± 3.8 years) were assigned to four groups (double placebo [placebo], BCAA + placebo, placebo + taurine, and BCAA + taurine [combined]) and given a combination of 3.2g BCAA (or placebo) and 2g taurine (or placebo), three times a day, for two weeks prior to and three days after eccentric elbow flexor exercises. DOMS and muscle damage in the biceps brachii were subjectively and objectively evaluated using the visual analogue scale (VAS), upper arm circumference (CIR), and blood parameters (creatine kinase, lactate dehydrogenase [LDH], aldolase, and 8-hydroxydeoxyguanosine [8-OHdG]).  In the combined group, VAS and 8-OHdG two days after exercise, CIR two and three days after exercise and LDH from one to three days after exercise were significantly lower than the placebo group. The area under the curve from before exercise to four days later for CIR, LDH, and aldolase was also significantly lower in the combined group than in the placebo group.  This study indicates that taurine can be utilized for attenuating exercise-induced DOMS and muscle damage.

Glutamine is also an conditional essential amino acid.  It plays a  key role in the metabolism of major nutrients and is vital for the reconstruction of body protein and the metabolism of energy.  Glutamine is produced in the body where the majority of it is stored in muscles.  In fact, glutamine is the most the abundant amino acid in the body.  However, during heavy workouts the body’s glutamine levels can become depleted.  ENDUR3 includes 2.75g of glutamine to replenish those glutamine levels to prevent fatigue and to aid in muscle glycogen resynthesis during recovery.

In a double-blind study, fifteen male high-level runners participated in a study to examine the impact of glutamine and (or) carbohydrate supplementation on blood ammonia concentration, since high levels are a cause for both peripheral and central fatigue.  In pre-competitive training, the participants ran 120 min (approximately 34 km) outdoors on 4 occasions. On the first day, the 15 athletes ran without the use of supplements and blood samples were taken every 30 min. After that, each day for 4 days before the next 3 exercise trials, the athlete’s’ normal diets was supplemented in bolus with carbohydrate (1 g.kg(-1).d(-1)), glutamine (70 mg.kg(-1).d(-1)), or a combination of both.  During the control trial ammonia increased progressively to approximately 70% above rest concentration. Following supplementation, independent of treatment, ammonia was not different (p>0.05) for the first 60 min, but for the second hour it was lower than in the control (p<0.05). The study concluded that supplementation with glutamine in high-level, endurance athletes reduced the accumulation of blood ammonia during prolonged, strenuous exercise in a field situation.  These findings suggest glutamine can prevent both peripheral and central fatigue by its action on blood ammonia concentration.

A study was completed to determine the efficacy of glutamine in promoting whole body carbohydrate storage and muscle glycogen resynthesis during recovery from exhaustive exercise. Seven male participants completed a glycogen depleting exercise protocol, then consumed 330 ml of one of three drinks, 18.5% (wt/vol) glucose polymer solution, 8 g glutamine in 330 ml glucose polymer solution, or 8 g glutamine in 330 ml placebo, and also received a primed constant infusion of [1-13C]glucose for 2 h. Plasma glutamine concentration was increased after consumption of the glutamine drinks (0.7–1.1 mM, P , 0.05). In the second hour of recovery, whole body nonoxidative glucose disposal was increased by 25% after consumption of glutamine in addition to the glucose polymer (4.48 6 0.61 vs. 3.59 6 0.18 mmol/kg, P , 0.05).  These results indicate that ingestion of glutamine alone appeared to promote muscle glycogen resynthesis during recovery from exhaustive exercise, relative to that expected from studies in which no substrate was provided. The promotion of muscle glycogen synthesis by consumption of glucose polymer and of glutamine was not additive. However, the addition of glutamine to the glucose polymer drink resulted in a greater storage of carbohydrate in sites other than skeletal muscle, the most likely candidate being the liver.

Maximum Hydration Matrix

During extensive periods of strenuous exercise, fluid loss occurs which can lead to thermal stress, accelerated fatigue, impaired cognition and a reduction in exercise performance.  Re-hydration with water is never a bad idea.  However, DemiGods train hard and need more than just plain water for replenishment.  The body loses electrolytes through sweat and they should be replaced by drinking fluids that contain electrolytes to prevent loss of energy, fatigue and fluid imbalance. That mission is accomplished with the Optimized Hydration Blend with provides you with the full set of electrolytes; 300mg of Sodium Chloride (NaCL), 140mg of Potassium Gluconate, 80mg of Magnesium Aspartate and 160mg of Calcium Citrate.  We also included 500mg of coconut water to maintain hydration, prevent cramping and reduce fatigue.

Electrolytes are essentials minerals that the body needs to function.  These include sodium, potassium, chloride, calcium and magnesium.  Electrolytes direct water and nutrients to the areas in the body where it is needed most and maintains an optimal fluid balance or in other words they support hydration.  In fact, by consuming fluids with electrolytes your body will require less overall fluid intake compared to plain water which will help you avoid carrying excess weight while you train.  Electrolytes also help your muscles to contract and relax which prevents cramping.  In addition, electrolytes can provide energy by breaking down protein and absorbing nutrients.  Magnesium is specifically beneficial since it participates in the conversion of Adenosine Triphosphate (ATP) and can lower blood pressure, heart rate and oxygen intake.

In one study eight participants consumed plain water and eight participants consumed water plus an electrolyte additive during 15 hours of laborious activity (ie.wildfire suppression).  There were no differences between groups for body weight, drinking frequency, temperature data, activity, or urine-specific gravity (1.019 ± 0.007 to 1.023 ± 0.010 vs. 1.019 ± 0.005 to 1.024 ± 0.009 for water and water + electrolyte groups pre- and post-shift, respectively; P < .05).  The water group consumed more total fluid (main effect for treatment) than the water + electrolyte group (504 ± 472 vs. 285 ± 279 mL·h-¹ for the water and water + electrolyte groups, respectively; P < .05).  This study found the addition of an electrolyte mixture to plain water decreased the overall fluid consumption of the water + electrolyte group by 220 mL·h-¹ (3.3 L·d-¹). Supplementing water with electrolytes can reduce the amount of fluid necessary to consume and transport during extended activity. This can minimize carrying excessive weight, possibly reducing fatigue during extended exercise.

Coconut water is a naturally occurring substance that is rich in potassium, contains sodium, chloride and carbohydrates.  It can be considered a viable alternative to water and sports drinks for those who need to ENDUR3 through gruelling periods of exercise that often require carbohydrate and electrolytes.  An added benefit of coconut water is that it causes less nausea and stomach upset than plain water and typical sports drinks  Olympus Labs uses organic coconut water powder in ENDUR3, only the best for a DemiGod.

Ten healthy male subjects participated in a study to assess the effectiveness of plain water (PW), sports drink (SD), fresh young coconut water (CW) and sodium-enriched fresh young coconut water (SCW) on whole body rehydration (R) and plasma volume (PV) restoration after exercise-induced dehydration.  Following a running session at 65% of VO2max, subjects drank, in randomized order, PW, SD, CW or SCW in three boluses representing 50, 40 and 30% of the fluid lost at 0, 30, and 60 minutes, respectively. In all trials subjects were still somewhat dehydrated even after the rehydration period. Indexes of percent rehydration with PW, SD, CW and SCW were 58 +/- 2, 68 +/- 2, 65+/- 2 and 69 +/- 1%, respectively, with significantly better rehydration with SD and SCW. The rehydration indexes for SD and SCW were significantly lower than PW (p < 0.01). PV was restored to euhydration levels after 2 hours of rehydration with SD, CW and SCW but not with PW. The plasma glucose concentration were significantly higher when SD, CW and SCW were ingested. SCW was similar in sweetness to CW and SD but caused less nausea and stomach upset compared to SD and PW. The study concluded SCW was as good as ingesting a commercial sports drink for whole body rehydration after exercise-induced dehydration but with better fluid tolerance.

Maximum Amino Uptake & Ergogenic Matrix

EAAs are great, but is that enough for peak performance?  Certainly not for a DemiGod or DemiGoddess to ENDUR3 through their intense training sessions.  That is why we assembled a blend of performance enhancing ingredients to more effectively utilize the amino acid blend and supercharge your workout. There is 2g of HydroMax™ Glycerol and 1g Agmatine that can both help reduce fatigue and also improve endurance.  We also included 150mg of schisandra (standardized to 9% Schisandrins) for its ability to reduce stress, increase blood flow and stimulate glucose uptake.  The matrix is perfected with 100mg of (-)/-epicatechin which can enhance exercise and oxidative capacity and increase angiogenesis (creation of blood vessel formation).

Glycerol is a naturally occurring metabolite that has several benefits for athletes. It can prevent athletes and bodybuilders from becoming dehydrated during the course of strenuous aerobic or anaerobic exercise. Glycerol can also enhance exercise performance and endurance for bodybuilders because it can keep the body hyper-hydrated.  

It has been demonstrated in human studies that hyperhydration with glycerol combined with water is far superior to water alone, and significantly increased fluid retention by approximately 900mL over placebo.  Of course an endurance athlete can also drink an over exceeding amount of water, however this will only help the athlete reach a temporary state of hyperhydration due to the fall in osmolarity causing the kidneys to remove the excess water and causing the athlete to make frequent trips to the bathroom.

ENDUR3 uses, HydroMax™ Glycerol, highly concentrated powdered glycerol which yields 65% free molecular glycerol.  Regular Glycerol Monostearate (GMS) contains only 5-12% free molecular glycerol, which is nowhere near the acceptable or effective range of glycerol supplementation.  This means 1.5 g of HydroMax™ Glycerol in ENDUR3 equates to more than 8g of the standard GMS found on the market!

Glycerol’s osmoprotective solute quality can be used to improve physical endurance. The reduction in serum blood osmolality and the effects on ionic gradients caused by glycerol ingestion delay fatigue; therefore, endurance and athletic performance are improved.

In a study assessing the effect of glycerol supplementation on aerobic and anaerobic performance of athletes and sedentary subjects, 40 volunteers were divided into four groups.  The first was the placebo group (S), which only consumed water; the second group (GS) consumed glycerol followed by water.  Neither of these groups did any exercise for 20 days. The third and fourth groups consisted of the exercise group subjects; they were required to perform a 20-m shuttle run test every day for 20 days. The third group’s subjects, the exercise placebo (E), only consumed water. The last group (GE) consumed glycerol followed by water. The glycerol supplement was found to have an increasing effect on aerobic and anaerobic performance in GS, E and GE. A similar effect was found for the covered distances and time in the same groups.

Agmatine ((4-aminobutyl)guanidine) is known as the ultimate nitric oxide enhancing ingredient. It is a byproduct of arginine produced through a process called decarboxylation. Agmatine is similar to arginine but with the carboxylic acid end removed.  Agmatine’s primary benefit occurs when it interacts with endothelial cells. These cells form a thin layer along blood vessel walls. As agmatine molecules pass through blood vessels, they attach to receptors on endothelial cell membranes. In response, endothelial cells make nitric oxide, a gas that dilates blood vessels.

A study was conducted examining the effects of agmatine on endothelial cell function by using cultured bovine pulmonary artery endothelial cells. Agmatine stimulated nitrite production three-fold above basal nitrite formation by endothelial cells.  This result suggests agmatine can act directly on endothelial cells to increase the synthesis of nitric oxide.

Agmatine has also been shown to increase glucose uptake in animal studies.  Agmatine acts upon the adrenal glands via the imidazoline receptors to release beta-endorphin and disposes glucose from the blood into skeletal muscle tissue.  Although this has not been replicated in human trials, agmatine is widely used by bodybuilders as a nutrient partitioning supplement.

Schisandra Chinensis berry (Schisandra) is an adaptogen herb.  The adaptogenic properties of Schisandra Chinensis were first exposed through a large number of pharmacological and clinical studies carried out by Russian scientists.  The results were impressive enough for them to include Schisandra Chinensis in editions of the National Pharmacopoeia of the USSR and in the State Register of Drugs.  Schisandra is particularly adept as a stress reducer which leads to an anti-catabolic effect.  In healthy subjects, Schisandra increases endurance and accuracy of movement, mental performance and working capacity.  It also generates alterations in the basal levels of nitric oxide and cortisol in blood and saliva with subsequent effects on the blood cells, vessels and the central nervous system.

In a placebo controlled double blind study, standardized extracts of Schisandra Chinensis and Bryonia alba roots were applied to several groups of athletes.  After treatment with the adaptogen, heavy physical exercise did not increase salivary NO and cortisol in athletes, whereas athletes treated with placebo and engaged in heavy physical exercise had increased salivary NO.  These results confirm the stress protective effect of Schisandra and its benefits for athletes engaged in heavy physical exercise.

A clinical study was conducted to investigate the effect of liver protective herbal materials, Sesamin combined with the extract of Schisandra Chinensis berry (Schisandra) for its potential to improve blood flow in humans.  Ten healthy male subjects with mean age of 51 years consumed 2 tablets of sesamin-schisandra combination (SCH) twice daily for two weeks. Each SCH tablet contained 2.5 mg sesamin, 65 mg schisandra extract and 3.75 IU Vitamin E.  Following continuous intake of Schisandra for 2 weeks the fasting blood passage time was reduced by 9.0% and 9.7% at week 1 and week 2, respectively. The baseline blood passage time prior to intake of SCH tablets was 53.5±3.5 sec as compared to 47.2±3.2 sec and 47.2±1.7 sec at Week 1 and Week 2. Although this change was not statistically significant, there was a sustained decreasing trend in blood passage time during 2 weeks of Schisandra administration (Figure 5).  These results indicate the combined administration of sesamin with schisandra extract could improve blood fluidity after 1 week of oral intake and this effect was sustained up to 2 weeks.

Fructus Schisandrae, the fruit of Schisandra chinensis, has been traditionally used as a hypoglycemic agent in Asia and its extracts have been shown to improve insulin-stimulated glucose uptake in cell-based assays in previous studies.  In an animal study, a fraction of Fructus Schisandrae, the fruit of Schisandra Chinensis, have been shown to improve glucose homeostasis by increasing glucose disposal rates and enhancing hepatic insulin sensitivity by working as a PPAR-γ agonist.  

Epicatechin is a flavanol found in cocoa seeds and green tea that has been found to act as a myostatin inhibitor.  Myostatin is a protein that inhibits muscle cell growth and differentiation, so blocking its action would be beneficial to those seeking additional muscle mass.  However its inclusion in ENDUR3 is primarily for its ability to enhance exercise and oxidative capacity and increase angiogenesis.

In animal studies (-)/-epicatechin has been shown lower blood pressure.  This characteristic seems to be replicated in humans based on anecdotal feedback from users of EP1C UNLEASHED.  In addition, those who have tried EP1C UNLEASHED, phytofused -epi, know first hand how great an ingredient (-)/-epicatechin is for strength and endurance.  Since the -epi in ENDUR3 is not phytofused you won’t quite get quite the same strength and endurance as you would from phytofused -epi.  But that’s okay because you should be stacking ENDUR3 with EP1C UNLEASHED, so the benefits will be additive.
In a pilot trial with (-)/-epicatechin, human subjects received either a single dose (n = 9) of 50, 100 or 200 mg or multiple doses (n = 8) of 50 mg daily (q.d.) or twice daily (b.i.d) for 5 days. In the q.d. and b.i.d. groups, blood samples were analyzed for NO surrogates and follistatin levels as well as, platelet mitochondrial complexes I, V and citrate synthase activity levels.  On day 5, in the q.d. and b.i.d. groups, there were significant increases in plasma nitrite of 30% and 17%, respectively. In the q.d. group on day 5 vs. day 1, platelet mitochondrial complexes I, IV and citrate synthase activities demonstrated a significant increase of ∼92, 62 and 8%, respectively. Average day 5 follistatin AUC levels were ∼2.5 fold higher vs. day 1 AUC levels in the b.i.d. group. There were no adverse effects observed from consumption of (−)-EPI therefore it was deemed safe to use.  These findings suggest that (−)-EPI can elicit increases in NO metabolites, mitochondrial enzyme function and plasma follistatin levels as reported in other studies.
