Maximum Endurance Matrix:

In fact, ATP production during weight-lifting or anaerobic events, such as high-intensity repeated, short but intense bursts of activity are largely fueled by anaerobic respiration and through another mechanism of action known as phosphorylation. Phosphorylation is how creatine exerts its potent ATP generating effects and it has been hypothesized that COP elicits its results via the same mechanism or by acting as an intracellular buffer.

Creatinol-O-Phosphate is a synthetic analogue of creatine, but it should not be confused with creatine as it does not carry the same benefits.  It is actually metabolized as creatinol in the body once it loses the phosphorous group.  It is a relatively new ingredient designed to aid in strength, performance, and recovery.  Although COP has been shown to increase ATP like creatine, it has a higher rate of absorption in muscle tissue.  This will result in greater contractility of muscles meaning users are able to carry out a longer anabolic load.  Punishing your muscles for a longer period of time will help users gain more strength and size!  COP has been proven to possess anti-ischemic and anti-arrhythmic activities associated with improved ionic balance and heart performance.

In a study of patients with acute myocardial infarction, the levels of six serum enzymes were investigated following supplementation with 3 g of COP.  After 3 days, the six enzymes all decreased between 33% and 49% in the COP group as compared with the placebo group, reaching a degree of statistical significance of less than 0.01. The decrease in these enzymes was attributed to the protective action of COP on cell membrane permeability, in agreement with a series of previous studies, and suggests COP has a cardioprotective effect.

ElevATP® is a patented ingredient consisting of a proprietary blend of plant bio-inorganic trace minerals and a polyphenol-rich apple extract.  ElevATP® will increase levels of ATP in human blood following ingestion.  I know what you are thinking, oral Adenosine 5’-Triphosphate DiSodium (ATP) is already widely available.  While that is true, ElevATP® seeks to remedy several issues with supplementation with oral ATP; its poor bioavailability and its tendency to increase reactive oxygen species (ROS) and lactic acid production.

In a double blind, placebo-controlled study, 18 generally healthy adults were given either a 150 mg dose of ElevATP® or 50 mg of an encapsulated silica oxide (placebo).  The researchers measured an acute increase in blood levels of ATP by 64% (P=0.02) when compared to the placebo group.  In addition, the ROS and lactate level in the supplemented group were unchanged.

A second study with 20 healthy subjects who had fasted overnight (12-16 hours) and were administered a single, 150 mg serving dose of ElevATP® also showed an increase in blood levels of ATP.  The average levels of the control group were measured as 40% over baseline levels at 60 minutes and 28% over baseline levels at 120 minutes. Similar to the first study, ATP levels in cell-free plasma and lactate levels remained unchanged.

Incredible Pump Matrix:

If that endurance matrix wasn’t powerful enough on its own, the pump matrix in CONQU3R Unleashed is composed of four ingredients: 4 g of L-citrulline, 2 g of HydroMax™ Glycerol, 1.5 g Nitrosigine™ and 1 g Agmatine that help reduce fatigue and also improve endurance.  However, these four ingredients were primarily selected for their ability to increase blood flow.  The pump effect achieved through the delivery of sufficient blood and oxygen to your muscles will ultimately lead to muscle growth.  Of course CONQU3R Unleashed is designed with clinically effective doses to induce an insane pump, like nothing you have ever experienced.

L-citrulline increases arginine and ornithine plasma content, resulting in a spike in nitric oxide levels which gives you that sought after pump effect.  But wait, if L-citrulline will increase plasma levels of arginine and ornithine why not just supplement with L-arginine and L-ornithine directly?  The problem is that L-arginine and L-ornithine are not absorbed by the body as well as L-citrulline.  You would need upwards of 10 g or more which may cause GI distress.

In a study in healthy males, L-citrulline dose-dependently increased AUC and Cmax of plasma L-arginine concentration more effectively than L-arginine (P < 0.01). Moreover, urinary nitrate and cGMP were increased from 92 ± 10 to 125 ± 15 µmol mmol−1 creatinine (P = 0.01, 95% CI 8, 58) and from 38 ± 3.3 to 50 ± 6.7 nmol mmol−1 creatinine (P = 0.04, 95% CI 0.4, 24), respectively.

Glycerol is a naturally occurring metabolite that has several benefits for athletes. It can prevent athletes and bodybuilders from becoming dehydrated during the course of strenuous aerobic or anaerobic exercise. Glycerol can also enhance exercise performance and endurance for bodybuilders because it can keep the body hyper-hydrated.

It has been demonstrated in human studies that hyperhydration with glycerol combined with water is far superior to water alone, and significantly increased fluid retention by approximately 900 mL over placebo.  Of course an endurance athlete can also drink an over exceeding amount of water, however this will only help the athlete reach a temporary state of hyperhydration due to the fall in osmolarity causing the kidneys to remove the excess water and causing the athlete to make frequent trips to the bathroom.

Regular Glycerol Monostearate (GMS) contains only 5-12% free molecular glycerol, which is nowhere near the acceptable or effective range of glycerol supplementation.  CONQU3R Unleashed uses, HydroMax™ Glycerol, highly concentrated powdered glycerol which yields 65% free molecular glycerol. Glycerol’s osmoprotective solute quality can be used to improve physical endurance. The reduction in serum blood osmolality and the effects on ionic gradients caused by glycerol ingestion delay fatigue; therefore, endurance and athletic performance are improved.

In a study assessing the effect of glycerol supplementation on aerobic and anaerobic performance of athletes and sedentary subjects, 40 volunteers were divided into four groups.  The first was the placebo group (S), which only consumed water; the second group (GS) consumed glycerol followed by water.  Neither of these groups did any exercise for 20 days. The third and fourth groups consisted of the exercise group subjects; they were required to perform a 20-m shuttle run test every day for 20 days. The third group’s subjects, the exercise placebo (E), only consumed water. The last group (GE) consumed glycerol followed by water. The glycerol supplement was found to have an increasing effect on aerobic and anaerobic performance in GS, E and GE. A similar effect was found for the covered distances and time in the same groups.

Nitrosigine™ is a patented complex of bonded arginine silicate scientifically designed to significantly boost nitric oxide (NO) levels.  We previously reviewed the positive effect from arginine on NO.  Unlike supplementing directly with L-arginine which could get  “messy,“ the Nitrosigine™ complex was shown in preclinical (animal) studies to be absorbed efficiently, raising plasma arginine and silicon levels. Silicon can strengthen and provide flexibility to your arterial walls, helping to increase blood flow.  Silicon is not well absorbed through dietary intake, but Nitrosigine™  was shown in preclinical studies to dramatically increase silicon absorption.

Another study found that a complex of arginine-silicate in an efficiently absorbed form, improves vascular function and reduces nephropathy in an animal model of insulin resistance and cardiovascular disease whereas arginine-hcl was not effective.

In a study in healthy males, supplementation with Nitrosigine™ caused an acute increase in arginine, as quick as 30 minutes.  The study also found a significant increase in arginine blood levels for up to 3 hours and silicon levels for up to 1.5 hours.  Furthermore, the data showed an increased trend in baseline levels of arginine after subsequent use.  So drink CONQU3R Unleashed 30 minutes before your workout, get a crazy pump for the whole duration and then get a better pump next workout.  Sign me up!

Agmatine ((4-aminobutyl)guanidine) is known as the ultimate nitric oxide enhancing ingredient. It is a byproduct of arginine produced through a process called decarboxylation. Agmatine is similar to arginine but with the carboxylic acid end removed.  Agmatine’s primary benefit occurs when it interacts with endothelial cells. These cells form a thin layer along blood vessel walls. As agmatine molecules pass through blood vessels, they attach to receptors on endothelial cell membranes. In response, endothelial cells make nitric oxide, a gas that dilates blood vessels.

A study was conducted examining the effects of agmatine on endothelial cell function by using cultured bovine pulmonary artery endothelial cells. Agmatine stimulated nitrite production three-fold above basal nitrite formation by endothelial cells.  This result suggests agmatine can act directly on endothelial cells to increase the synthesis of nitric oxide.

Maximum Focus Matrix:

In a randomized, placebo-controlled, crossover design, seven men with at least two years of resistance training experience ingested 600 mg A-GPC (as AlphaSize®) or a placebo 90-minutes prior to completing 6 sets × 10 repetitions of Smith Machine squats at 70% of their predetermined 1-repetition maximum.   Measurements were taken 30 minutes post-exercise and then the subjects performed three sets of bench press throws at 50% of their predetermined 1-repetition maximum to assess peak force, peak power, and rate of force development. All trials were performed after an overnight fast, a 48-hour abstention from intense exercise, and during the same time of day to minimize diurnal variation.  Compared to baseline (pre) values, peak GH increased 44-fold during A-GPC (from 0.19 ± 0.06 to 8.4 ± 2.1 ng/mL) vs. 2.6-fold during placebo (from 1.9 ± 0.8 to 5.0 ± 4.8 ng/mL, P < 0.03) (Figure 3). Peak bench press force was 14% greater in A-GPC (933 ± 89 N) vs. placebo (818 ± 77 N, P < 0.02).

Research has also shown it should improve your ability to lose fat over time due to an increase in hepatic (liver) fat oxidation.  In the same study, a significant increase in growth hormone was measured after 60 minutes of ingestion, indicating an acute response making it beneficial for supplementation pre-workout.

Acetyl-L-Carnitine, otherwise known as ALCAR, is a carnitine molecule bound to an acetyl group. While L-carnitine is known for its fat loss benefits, ALCAR is known for its neurological benefits.  It is included in CONQU3R Unleashed for its ability to increase alertness and reduce mental fatigue.  It is such a potent brain booster that treatment with carnitine significantly decreased attention problems and aggressive behavior in boys with ADHD.

In a double-blind, placebo-controlled study, patients with chronic fatigue syndrome were given 2 g per day of ALCAR.  After just two weeks, 52% of patients reported a significant reduction in mental fatigue.

In another double-blind, placebo-controlled study, 30 elderly patients were given 2 g per day of ALCAR for two months.  The group that was given ALCAR showed a statistically significant improvement in the behavioral scales, in the memory tests, in the attention barrage test and in the Verbal Fluency test.

Maximum Adaptogen Matrix:

Adaptogens are substances which normalize body functions and increase resistance to stress. Recreationally trained males were given 800 mg per day of Eleutherococcus senticosus (ES) for 8 weeks.  The examined physiological variables included endurance time, maximal heart rate during exhaustion exercise, VO2, rating of perceived exertion and respiratory exchange ratio after the subjects cycled at 75% VO2 peak until exhaustion. The VO2 peak of the subjects elevated 12% (P < 0.05), endurance time improved 23% (P < 0.05) and peak heart rate increased 4% (P < 0.05).  In addition, at 30 min of 75% VO2 peak cycling, the production of plasma FFA was increased and the glucose level was decreased both significantly (P < 0.05) over 8-week ES supplementation. This study demonstrates long term supplementation of ES can enhance endurance capacity, elevate cardiovascular functions and alter the metabolism for sparing glycogen in active individuals.

Rhodiola Rosea Extract is one of the most effective adaptogen compounds for its significant ability to mitigate the undesirable effects of prolonged exercise, like physical exhaustion and fatigue.  The end result is an increase in endurance, and with the combination of the endurance matrix in CONQU3R Unleashed Stim-free you will feel like you can stay in the gym all day.  Rhodiola Rosea has also been shown to possess antioxidant properties which will moderate undue stress to your body from an intense workout.

In a study with 24 healthy volunteers, 12 male and 12 female, were given a dose of 100 mg Rhodiola Rosea.  After just 4 days of supplementation a significant increase in time to exhaustion, VO2, VCO2, peak O2 output, and peak CO2 output was observed.

Another study, with 22 professional rowers, that were given 100 mg of Rhodiola Rosea twice daily for a 4 week period found a significant increase in total plasma antioxidant capacity and a decrease in superoxide dismutase (SOD) activity.  SOD is an enzyme produced as a result of oxygen metabolism (ie. intensive activity) and can lead to cell damage if not mitigated.  These results suggest that continuous supplementation of Rhodiola Rosea can improve endurance.

The adaptogenic properties of Schisandra Chinensis were first exposed through a large number of pharmacological and clinical studies carried out by Russian scientists.  The results were impressive enough for them to include Schisandra Chinensis in editions of the National Pharmacopoeia of the USSR and in the State Register of Drugs.  Schisandra is particularly adept as stress reducer, but also has several other benefits.  In healthy subjects, Schisandra increases endurance and accuracy of movement, mental performance and working capacity, and generates alterations in the basal levels of nitric oxide and cortisol in blood and saliva with subsequent effects on the blood cells, vessels and central nervous system.

Several groups of athletes in a placebo-controlled, double-blind study were given Schisandra Chinensis prior to heavy physical exercise.  After treatment with the adaptogen, heavy physical exercise did not increase salivary nitric oxide (NO) and cortisol in athletes, whereas athletes treated with placebo increased salivary NO.

In a double-blind, placebo-controlled, randomized trial, forty healthy females aged between 20-68 years were given a standardized fixed combination of Rhodiola Rosea L., Schisandra Chinensis and Eleutherococcus senticosus extracts.  The effects of the extract were  measured prior to treatment and two hours after treatment using the d2 Test of Attention. The subjects in the control group quickly (two hours after supplementation) gained improved attention and increased speed and accuracy during stressful cognitive tasks, in comparison to placebo.  CONQU3R Unleashed Stim-free just so happens to includes all three adaptogens.

The inclusion of dimethylaminoethanol (DMAE) is primarily for its ability to increase levels of acetylcholine resulting in an increase in attention and focus. However, DMAE is known to have much better transportation to the brain than choline and can result in a higher level of cholinergic activity.

In a randomized, group-parallel, double-blind, placebo-controlled study, 40 males and 40 females ingested a vitamin-mineral drug combination containing dimethylaminoethanol (DMAE) or placebo.  Measurements of their brain’s electrical reaction were taken during presentation of 5 videoclips of 7 min duration followed by 3 minutes pause. This procedure was repeated after 6 and 12 weeks of daily supplementation with DMAE.  The EEG analysis showed decreases in theta and alpha1 electrical power which have been associated with increased vigilance and attention.  A subsequent questionnaire given to the subjects revealed a better mood for the active drug group thus corroborating the results from EEG analysis. The study concluded DMAE can be interpreted to induce a psychophysiological state of better feeling of wellbeing on both levels of analysis mood and electrical pattern of brain activity.

Maximum mTOR Igniting BCAA & EAA Matrix

Amino acids are organic compounds that combine to form proteins.  They are commonly referred to as the building blocks of protein.  There are 3 classes of amino acids; non-essential, essential and conditional.   Non-essential amino acids are naturally produced in the body whereas essential amino acids are not synthesized by the body and must come from diet and/or supplementation.  There are 9 essential amino acids (EAAs) which promote muscle protein synthesis (MPS).  Of course the Maximum mTOR Igniting BCAA & EAA Matrix includes all 9 EAAs plus l-tyrosine.  L- tyrosine has been found to have an acute effect in preventing a decline in cognitive function in response to physical stress.  

Among the nine EAAs there are three aminos known as branched chain amino acids (BCAAs); leucine, isoleucine and valine.  BCAAs have the greatest impact on MPS and fatigue reduction.  Skeletal muscle is a major protein source for catabolism and BCAAs are metabolized during this process so it is important that you are getting an adequate amount in your diet.  For this reason the Crazy 10 Anabolic Amino Blend contains a proportional amount of BCAAs with 5g leucine, 1.5g isoleucine and 1.5g valine.

A clinical trial was conducted to determine the effect of EAA supplementation with a higher amount of leucine content on postexercise skeletal muscle metabolism and MPS.  Study participants consumed an EAA drink with equivalent amounts of dietary nitrogen (10g) during exercise.  However, one group consumed an EAA drink with 3.5g leucine content (L-EAA) while the other group consumed an EAA drink with 1.87g leucine content (EAA). leucine.  MPS was 33% greater (P < 0.05) after consumption of L-EAA (0.08 ± 0.01%/h) than after consumption of EAA (0.06 ± 0.01%/h). Whole-body protein breakdown and synthesis were lower (P < 0.05) and oxidation was greater (P < 0.05) after consumption of L-EAA than after consumption of EAA.  These findings indicate that consuming an EAA supplement with a higher leucine content during endurance exercise elicits a greater MPS response during recovery.  ENDUR3 goes even further than the studied dose of 3.5g with a massive dose of 5g of leucine to maximize MPS.

As the following study demonstrates, supplementation with EAAs in the absence of regular exercise is still beneficial.  14 elderly women (68 +/- 2 yr) participated in a randomized, double-blinded, placebo-controlled study to assess EAA supplementation on postabsorptive muscle protein fractional synthesis rate (FSR), lean body mass (LBM), and one-repetition maximum muscle strength, and androgen receptor and IGF-I muscle protein expression.  The supplemented group (SUP) consumed 15g EAA/d and the control group ingested a placebo for a period of 3 months.  Ingestion of 7.5 g EAA acutely stimulated FSR in both groups at 0 months (P < 0.05). Basal FSR at 3 months was increased in SUP only. The magnitude of the acute response to EAA was unaltered after 3 months in SUP. LBM increased in SUP only (P < 0.05). One-repetition maximum strength remained unchanged in both groups. Basal IGF-I protein expression increased in SUP after 3 months (P = 0.05), with no changes in androgen receptor or total and phosphorylated Akt, mTOR, S6 kinase, and 4E-binding protein.  This study shows that supplementation with EAA has an acute anabolic response and is maintained over time and can improve LBM.

Fatigue Destroying Matrix

It is natural to feel sore and fatigued when you are pushing yourself to the limits and gaining muscle.  DemiGods have no tolerance for fatigue, we must ENDUR3 so we can go harder during the next training session.  Say goodbye to muscle soreness with the Fatigue Destroying Matrix.  It includes 2.75g of L-glutamine and 1g of taurine to minimize muscle soreness and fatigue.

Taurine is an amino acid that has a sulfonic acid chain attached to it, in contrast to other amino acids that have a carboxyl acid chain.  It is classified as an conditional essential amino acid, meaning it is essential only during periods of diseases or physical stress such as strenuous exercise.  However, your body not produce enough taurine on its own so it is important to obtain the amino sulfonic acid through diet and/or supplementation. Taurine has several benefits including restoring insulin sensitivity, mitigating diabetic complications, reversing cardiovascular disease factors, preventing and treating fatty liver disease.  Most pertinent for athletes is its ability to reduce muscle soreness and cramping.

Thirty-six untrained male subjects (22.5 ± 3.8 years) were assigned to four groups (double placebo [placebo], BCAA + placebo, placebo + taurine, and BCAA + taurine [combined]) and given a combination of 3.2g BCAA (or placebo) and 2g taurine (or placebo), three times a day, for two weeks prior to and three days after eccentric elbow flexor exercises. DOMS and muscle damage in the biceps brachii were subjectively and objectively evaluated using the visual analogue scale (VAS), upper arm circumference (CIR), and blood parameters (creatine kinase, lactate dehydrogenase [LDH], aldolase, and 8-hydroxydeoxyguanosine [8-OHdG]).  In the combined group, VAS and 8-OHdG two days after exercise, CIR two and three days after exercise and LDH from one to three days after exercise were significantly lower than the placebo group. The area under the curve from before exercise to four days later for CIR, LDH, and aldolase was also significantly lower in the combined group than in the placebo group.  This study indicates that taurine can be utilized for attenuating exercise-induced DOMS and muscle damage.

Glutamine is also an conditional essential amino acid.  It plays a  key role in the metabolism of major nutrients and is vital for the reconstruction of body protein and the metabolism of energy.  Glutamine is produced in the body where the majority of it is stored in muscles.  In fact, glutamine is the most the abundant amino acid in the body.  However, during heavy workouts the body’s glutamine levels can become depleted.  ENDUR3 includes 2.75g of glutamine to replenish those glutamine levels to prevent fatigue and to aid in muscle glycogen resynthesis during recovery.

In a double-blind study, fifteen male high-level runners participated in a study to examine the impact of glutamine and (or) carbohydrate supplementation on blood ammonia concentration, since high levels are a cause for both peripheral and central fatigue.  In pre-competitive training, the participants ran 120 min (approximately 34 km) outdoors on 4 occasions. On the first day, the 15 athletes ran without the use of supplements and blood samples were taken every 30 min. After that, each day for 4 days before the next 3 exercise trials, the athlete’s’ normal diets was supplemented in bolus with carbohydrate (1 g.kg(-1).d(-1)), glutamine (70 mg.kg(-1).d(-1)), or a combination of both.  During the control trial ammonia increased progressively to approximately 70% above rest concentration. Following supplementation, independent of treatment, ammonia was not different (p>0.05) for the first 60 min, but for the second hour it was lower than in the control (p<0.05). The study concluded that supplementation with glutamine in high-level, endurance athletes reduced the accumulation of blood ammonia during prolonged, strenuous exercise in a field situation.  These findings suggest glutamine can prevent both peripheral and central fatigue by its action on blood ammonia concentration.

A study was completed to determine the efficacy of glutamine in promoting whole body carbohydrate storage and muscle glycogen resynthesis during recovery from exhaustive exercise. Seven male participants completed a glycogen depleting exercise protocol, then consumed 330 ml of one of three drinks, 18.5% (wt/vol) glucose polymer solution, 8 g glutamine in 330 ml glucose polymer solution, or 8 g glutamine in 330 ml placebo, and also received a primed constant infusion of [1-13C]glucose for 2 h. Plasma glutamine concentration was increased after consumption of the glutamine drinks (0.7–1.1 mM, P , 0.05). In the second hour of recovery, whole body nonoxidative glucose disposal was increased by 25% after consumption of glutamine in addition to the glucose polymer (4.48 6 0.61 vs. 3.59 6 0.18 mmol/kg, P , 0.05).  These results indicate that ingestion of glutamine alone appeared to promote muscle glycogen resynthesis during recovery from exhaustive exercise, relative to that expected from studies in which no substrate was provided. The promotion of muscle glycogen synthesis by consumption of glucose polymer and of glutamine was not additive. However, the addition of glutamine to the glucose polymer drink resulted in a greater storage of carbohydrate in sites other than skeletal muscle, the most likely candidate being the liver.

Maximum Hydration Matrix

During extensive periods of strenuous exercise, fluid loss occurs which can lead to thermal stress, accelerated fatigue, impaired cognition and a reduction in exercise performance.  Re-hydration with water is never a bad idea.  However, DemiGods train hard and need more than just plain water for replenishment.  The body loses electrolytes through sweat and they should be replaced by drinking fluids that contain electrolytes to prevent loss of energy, fatigue and fluid imbalance. That mission is accomplished with the Optimized Hydration Blend with provides you with the full set of electrolytes; 300mg of Sodium Chloride (NaCL), 140mg of Potassium Gluconate, 80mg of Magnesium Aspartate and 160mg of Calcium Citrate.  We also included 500mg of coconut water to maintain hydration, prevent cramping and reduce fatigue.

Electrolytes are essentials minerals that the body needs to function.  These include sodium, potassium, chloride, calcium and magnesium.  Electrolytes direct water and nutrients to the areas in the body where it is needed most and maintains an optimal fluid balance or in other words they support hydration.  In fact, by consuming fluids with electrolytes your body will require less overall fluid intake compared to plain water which will help you avoid carrying excess weight while you train.  Electrolytes also help your muscles to contract and relax which prevents cramping.  In addition, electrolytes can provide energy by breaking down protein and absorbing nutrients.  Magnesium is specifically beneficial since it participates in the conversion of Adenosine Triphosphate (ATP) and can lower blood pressure, heart rate and oxygen intake.

In one study eight participants consumed plain water and eight participants consumed water plus an electrolyte additive during 15 hours of laborious activity (ie.wildfire suppression).  There were no differences between groups for body weight, drinking frequency, temperature data, activity, or urine-specific gravity (1.019 ± 0.007 to 1.023 ± 0.010 vs. 1.019 ± 0.005 to 1.024 ± 0.009 for water and water + electrolyte groups pre- and post-shift, respectively; P < .05).  The water group consumed more total fluid (main effect for treatment) than the water + electrolyte group (504 ± 472 vs. 285 ± 279 mL·h-¹ for the water and water + electrolyte groups, respectively; P < .05).  This study found the addition of an electrolyte mixture to plain water decreased the overall fluid consumption of the water + electrolyte group by 220 mL·h-¹ (3.3 L·d-¹). Supplementing water with electrolytes can reduce the amount of fluid necessary to consume and transport during extended activity. This can minimize carrying excessive weight, possibly reducing fatigue during extended exercise.

Coconut water is a naturally occurring substance that is rich in potassium, contains sodium, chloride and carbohydrates.  It can be considered a viable alternative to water and sports drinks for those who need to ENDUR3 through gruelling periods of exercise that often require carbohydrate and electrolytes.  An added benefit of coconut water is that it causes less nausea and stomach upset than plain water and typical sports drinks  Olympus Labs uses organic coconut water powder in ENDUR3, only the best for a DemiGod.

Ten healthy male subjects participated in a study to assess the effectiveness of plain water (PW), sports drink (SD), fresh young coconut water (CW) and sodium-enriched fresh young coconut water (SCW) on whole body rehydration (R) and plasma volume (PV) restoration after exercise-induced dehydration.  Following a running session at 65% of VO2max, subjects drank, in randomized order, PW, SD, CW or SCW in three boluses representing 50, 40 and 30% of the fluid lost at 0, 30, and 60 minutes, respectively. In all trials subjects were still somewhat dehydrated even after the rehydration period. Indexes of percent rehydration with PW, SD, CW and SCW were 58 +/- 2, 68 +/- 2, 65+/- 2 and 69 +/- 1%, respectively, with significantly better rehydration with SD and SCW. The rehydration indexes for SD and SCW were significantly lower than PW (p < 0.01). PV was restored to euhydration levels after 2 hours of rehydration with SD, CW and SCW but not with PW. The plasma glucose concentration were significantly higher when SD, CW and SCW were ingested. SCW was similar in sweetness to CW and SD but caused less nausea and stomach upset compared to SD and PW. The study concluded SCW was as good as ingesting a commercial sports drink for whole body rehydration after exercise-induced dehydration but with better fluid tolerance.

Maximum Amino Uptake & Ergogenic Matrix

EAAs are great, but is that enough for peak performance?  Certainly not for a DemiGod or DemiGoddess to ENDUR3 through their intense training sessions.  That is why we assembled a blend of performance enhancing ingredients to more effectively utilize the amino acid blend and supercharge your workout. There is 2g of HydroMax™ Glycerol and 1g Agmatine that can both help reduce fatigue and also improve endurance.  We also included 150mg of schisandra (standardized to 9% Schisandrins) for its ability to reduce stress, increase blood flow and stimulate glucose uptake.  The matrix is perfected with 100mg of (-)/-epicatechin which can enhance exercise and oxidative capacity and increase angiogenesis (creation of blood vessel formation).

Glycerol is a naturally occurring metabolite that has several benefits for athletes. It can prevent athletes and bodybuilders from becoming dehydrated during the course of strenuous aerobic or anaerobic exercise. Glycerol can also enhance exercise performance and endurance for bodybuilders because it can keep the body hyper-hydrated.  

It has been demonstrated in human studies that hyperhydration with glycerol combined with water is far superior to water alone, and significantly increased fluid retention by approximately 900mL over placebo.  Of course an endurance athlete can also drink an over exceeding amount of water, however this will only help the athlete reach a temporary state of hyperhydration due to the fall in osmolarity causing the kidneys to remove the excess water and causing the athlete to make frequent trips to the bathroom.

ENDUR3 uses, HydroMax™ Glycerol, highly concentrated powdered glycerol which yields 65% free molecular glycerol.  Regular Glycerol Monostearate (GMS) contains only 5-12% free molecular glycerol, which is nowhere near the acceptable or effective range of glycerol supplementation.  This means 1.5 g of HydroMax™ Glycerol in ENDUR3 equates to more than 8g of the standard GMS found on the market!

Glycerol’s osmoprotective solute quality can be used to improve physical endurance. The reduction in serum blood osmolality and the effects on ionic gradients caused by glycerol ingestion delay fatigue; therefore, endurance and athletic performance are improved.

In a study assessing the effect of glycerol supplementation on aerobic and anaerobic performance of athletes and sedentary subjects, 40 volunteers were divided into four groups.  The first was the placebo group (S), which only consumed water; the second group (GS) consumed glycerol followed by water.  Neither of these groups did any exercise for 20 days. The third and fourth groups consisted of the exercise group subjects; they were required to perform a 20-m shuttle run test every day for 20 days. The third group’s subjects, the exercise placebo (E), only consumed water. The last group (GE) consumed glycerol followed by water. The glycerol supplement was found to have an increasing effect on aerobic and anaerobic performance in GS, E and GE. A similar effect was found for the covered distances and time in the same groups.

Agmatine ((4-aminobutyl)guanidine) is known as the ultimate nitric oxide enhancing ingredient. It is a byproduct of arginine produced through a process called decarboxylation. Agmatine is similar to arginine but with the carboxylic acid end removed.  Agmatine’s primary benefit occurs when it interacts with endothelial cells. These cells form a thin layer along blood vessel walls. As agmatine molecules pass through blood vessels, they attach to receptors on endothelial cell membranes. In response, endothelial cells make nitric oxide, a gas that dilates blood vessels.

A study was conducted examining the effects of agmatine on endothelial cell function by using cultured bovine pulmonary artery endothelial cells. Agmatine stimulated nitrite production three-fold above basal nitrite formation by endothelial cells.  This result suggests agmatine can act directly on endothelial cells to increase the synthesis of nitric oxide.

Agmatine has also been shown to increase glucose uptake in animal studies.  Agmatine acts upon the adrenal glands via the imidazoline receptors to release beta-endorphin and disposes glucose from the blood into skeletal muscle tissue.  Although this has not been replicated in human trials, agmatine is widely used by bodybuilders as a nutrient partitioning supplement.

Schisandra Chinensis berry (Schisandra) is an adaptogen herb.  The adaptogenic properties of Schisandra Chinensis were first exposed through a large number of pharmacological and clinical studies carried out by Russian scientists.  The results were impressive enough for them to include Schisandra Chinensis in editions of the National Pharmacopoeia of the USSR and in the State Register of Drugs.  Schisandra is particularly adept as a stress reducer which leads to an anti-catabolic effect.  In healthy subjects, Schisandra increases endurance and accuracy of movement, mental performance and working capacity.  It also generates alterations in the basal levels of nitric oxide and cortisol in blood and saliva with subsequent effects on the blood cells, vessels and the central nervous system.

In a placebo controlled double blind study, standardized extracts of Schisandra Chinensis and Bryonia alba roots were applied to several groups of athletes.  After treatment with the adaptogen, heavy physical exercise did not increase salivary NO and cortisol in athletes, whereas athletes treated with placebo and engaged in heavy physical exercise had increased salivary NO.  These results confirm the stress protective effect of Schisandra and its benefits for athletes engaged in heavy physical exercise.

A clinical study was conducted to investigate the effect of liver protective herbal materials, Sesamin combined with the extract of Schisandra Chinensis berry (Schisandra) for its potential to improve blood flow in humans.  Ten healthy male subjects with mean age of 51 years consumed 2 tablets of sesamin-schisandra combination (SCH) twice daily for two weeks. Each SCH tablet contained 2.5 mg sesamin, 65 mg schisandra extract and 3.75 IU Vitamin E.  Following continuous intake of Schisandra for 2 weeks the fasting blood passage time was reduced by 9.0% and 9.7% at week 1 and week 2, respectively. The baseline blood passage time prior to intake of SCH tablets was 53.5±3.5 sec as compared to 47.2±3.2 sec and 47.2±1.7 sec at Week 1 and Week 2. Although this change was not statistically significant, there was a sustained decreasing trend in blood passage time during 2 weeks of Schisandra administration (Figure 5).  These results indicate the combined administration of sesamin with schisandra extract could improve blood fluidity after 1 week of oral intake and this effect was sustained up to 2 weeks.

Fructus Schisandrae, the fruit of Schisandra chinensis, has been traditionally used as a hypoglycemic agent in Asia and its extracts have been shown to improve insulin-stimulated glucose uptake in cell-based assays in previous studies.  In an animal study, a fraction of Fructus Schisandrae, the fruit of Schisandra Chinensis, have been shown to improve glucose homeostasis by increasing glucose disposal rates and enhancing hepatic insulin sensitivity by working as a PPAR-γ agonist.  

Epicatechin is a flavanol found in cocoa seeds and green tea that has been found to act as a myostatin inhibitor.  Myostatin is a protein that inhibits muscle cell growth and differentiation, so blocking its action would be beneficial to those seeking additional muscle mass.  However its inclusion in ENDUR3 is primarily for its ability to enhance exercise and oxidative capacity and increase angiogenesis.

In a pilot trial with (-)/-epicatechin, human subjects received either a single dose (n = 9) of 50, 100 or 200 mg or multiple doses (n = 8) of 50 mg daily (q.d.) or twice daily (b.i.d) for 5 days. In the q.d. and b.i.d. groups, blood samples were analyzed for NO surrogates and follistatin levels as well as, platelet mitochondrial complexes I, V and citrate synthase activity levels.  On day 5, in the q.d. and b.i.d. groups, there were significant increases in plasma nitrite of 30% and 17%, respectively. In the q.d. group on day 5 vs. day 1, platelet mitochondrial complexes I, IV and citrate synthase activities demonstrated a significant increase of ∼92, 62 and 8%, respectively. Average day 5 follistatin AUC levels were ∼2.5 fold higher vs. day 1 AUC levels in the b.i.d. group. There were no adverse effects observed from consumption of (−)-EPI therefore it was deemed safe to use.  These findings suggest that (−)-EPI can elicit increases in NO metabolites, mitochondrial enzyme function and plasma follistatin levels as reported in other studies.

MAXIMUM MTOR ACTIVATION

I am sure you have heard of mTOR because several companies make outlandish claims that their product will increase it significantly. Let’s briefly examine what it is and how it can benefit resistance trained individuals in order to develop some “BS radar”.  mTOR is an abbreviation for the mammalian target of rapamycin.  It is a protein kinase that is a well-known regulator of cellular growth. It has been widely established through several studies that the kinase activity of mTOR is required for mechanically-induced increases in skeletal muscle, protein synthesis and hypertrophy. Ultimately the goal of any training regimen is to grow muscle, therefore an increase in mTOR is definitely desirable.

Capsaicin is an active component from chili peppers, but we used the non-pungent form, Stearoyl Vanillylamide in TR1UMPH to prevent any associated burning sensation when it is ingested.  Capsaicin is a transient receptor potential vanilloid 1 (TRPV1) agonist. TRPV1 is a Ca2+ permeant non-selective cation channel expressed in a subpopulation of primary afferent neurons, conducting impulses inwards to the brain. It is included in TR1UMPH for its ability to induce increases in [Ca2+].  It has been established that the activation of mTOR and subsequent muscle hypertrophy requires a (TRPV1) mediated increase in intracellular calcium concentrations ([Ca2+]I).  Thus, calcium signaling is a crucial step for mechanical load-induced activation of mTOR.

In an animal model,  muscle cells were treated with the TRPV1 agonist capsaicin to analyze the effect on [Ca2+] using Fluo-4. [Ca2+] was increased by capsaicin in wild-type but not in TRPV1-null muscle fibers, indicating that the capsaicin induced increases in [Ca2+]i were mediated by TRPV1 (Fig.1A and 1B).  Since [Ca2+] regulates divergent intracellular signaling pathways, the capsaicin-induced intracellular signaling events were further analyzed. Similar to previous observations, intramuscular injection of capsaicin induced phosphorylation of p70S6K at Thr389, an mTOR- regulated phosphorylation site that is necessary for p70S6K activation in addition to phosphorylation of the downstream target S6. Capsaicin also induced phosphorylation of p70S6K at Thr421/Ser424 and mTOR at Ser2448 and Ser2481. In addition to p70S6K, capsaicin induced phosphorylation of Erk1/2 and p38 MAPK, but not Akt, AMPKα, or GSK3β.

Capsaicin can also improve lipolysis which is the decomposition of stored fat.  This process activates enzymes that increase production of Cyclic adenosine monophosphate (cAMP) which activates protein kinase A.

In a study with 10 healthy males, ingestion of 150mg of capsaicin ingestion (150mg) enhances substrate oxidation associated with thermogenic sympathetic activity as an energy metabolic modulator without causing prolongation of the cardiac OT interval during aerobic exercise in humans.  In conclusion, it may be considered that capsaicin consumption 1 h before low intensity exercise (50% VT(max)) is a valuable supplement for the treatment of individuals with hyperlipidemia and/or obesity because it improves lipolysis without any adverse effects on the cardiac depolarization and repolarization process.

Prevention of muscle atrophy is based on either inhibiting protein-degradation pathways, which are activated during atrophy, or activating protein-synthesis pathways, which induce muscle hypertrophy..  As we previously reviewed, Capsaicin achieves the latter.  Rhodiola Rosea Extract, a Scandinavian herb,  possesses a significant ability to reduce stress on the body, particularly exhaustion and fatigue, caused through physical exertion. Rhodiola Rosea has also been shown to possess antioxidant properties which will also moderate undue stress to your body from intense exercise.

24 healthy volunteers, 12 male and 12 female, were given a dose of 100mg Rhodiola Rosea.  After just 4 days of supplementation a significant increase in time to exhaustion, VO2, VCO2, peak O2 output, and peak CO2 output was observed.

Another study, with 22 professional rowers, that were given 100mg of Rhodiola Rosea twice daily for a 4 week period found a significant increase in total plasma antioxidant capacity and a decrease in superoxide dismutase (SOD) activity.  SOD is an enzyme produced as a result of oxygen metabolism (ie. intensive activity) and can lead to cell damage if not mitigated.  These results suggest that continuous supplementation of Rhodiola Rosea can improve endurance.

As previously mentioned TR1UMPH contains a purer extract of Rhodiola Rosea which yields 11% Salidroside.  That is no coincidence!  Olympus Labs invested the extra money to reap the benefits of this ingredient because DemiGods deserve the best!  Based on research conducted on human bone marrow samples, Salidroside was found to induce the phosphorlyation of Akt, mTOR and p70 S6 kinase.  In addition, Salidroside stimulates ERK1/2 which is associated with cell migration and capillary tube formation.  Further research has identified that Salidroside has the ability to increase the mRNA level of genes involved in the regulation of Bone Morphogenetic Protein (BMP) signaling pathway, including BMP2, BMP6 and BMP7.

BMPs include a large family of proteins within the TGF-beta superfamily of growth and differentiation factors. Members of the BMP family have been found to be key signaling molecules in embryogenesis, in species ranging from small flies to humans.  Research has shown BMP signaling is the fundamental signal for hypertrophy, specifically BMP7 which is the pathway that has the most influence on muscle growth.  BMP7 can stimulate osteoblast differentiation leading to formation of skeletal muscle.  Unfortunately the majority research on Salidroside is limited to testing on mice.  However, given the positive human data on Rhodiola Rosea and the preclinical data in humans it lends extra credibility to these findings.   

MAXIMUM ERGOGENIC IGNITION

An ergogenic aid can be broadly defined as a substance used for the purpose of enhancing athletic performance. There are several ergogenic aids that are so beneficial for high intensity exercise that they are considered “staples”. We included three such staples in TR1UMPH to elicit an increase in physical capacity and reduction in fatigue; 2.5g of creatine Hcl, 2.5g of betaine, and 2g of L-Carnitine L-Tartrate (LCLT).

Creatine is by far the most popular ergogenic supplement because it has proven to be effective by numerous studies.  In fact, it is likely the most studied performance enhancing supplement in the industry.  Of course we cannot review all of these studies so in order to be efficient; let’s review a meta-analysis.

One meta-analysis reviewed 16 studies involving healthy male and/or female subjects.  The summary difference in maximum weight lifted was 6.85kg (95% confidence interval [CI], 5.24-8.47) greater after creatine than placebo for bench press and 9.76 kg (95% CI, 3.37-16.15) greater for squats; there was no difference for arm curls. In 7 of 10 studies evaluating maximal weight lifted in young men (younger than 36 years) that engaged in resistance training there was no difference in cycle ergometer or isokinetic dynamometer performance.  The researchers concluded oral creatine supplementation combined with resistance training increases maximal weight lifted in young men. There is no evidence for improved performance in older individuals or women or for other types of strength and power exercises

A second meta-analysis of 96 English-language, peer-reviewed papers (100 studies), which included randomized group formation, a placebo control, and human subjects who were blinded to treatments was performed.  It compared the effects of creatine supplementation (CS) on effect size (ES) for body composition (BC) variables (mass and lean body mass), duration and intensity (< or = 30 s, [ATP-PCr = A]; 30-150 s [glycolysis = G]; >150 s, [oxidative phosphorylation = O]) of the exercise task, type of exercise task (single, repetitive, laboratory, field, upper-body, lower-body), CS duration (loading, maintenance), and subject characteristics (gender, training status). ES was greater for changes in lean body mass following short-term CS, repetitive-bout laboratory-based exercise tasks < or = 30 s (e.g., isometric, isokinetic, and isotonic resistance exercise), and upper-body exercise. There is no evidence in the literature of an effect of gender or training status on ES following CS

TR1UMPH uses creatine hydrochloride (HCL) which is creatine bound with hydrochloric acid. It’s turned into a basic creatine molecule by stomach acid so you get all the benefits of creatine without any potential side effects like upset stomach.  Creatine HCL is also more acidic (ie. lower PH) than creatine monohydrate which makes it more soluble and would suggest an increase in absorption. Since it is better absorbed you can supplement with a lower dosage compared to creatine monohydrate to receive the same strength, power and muscle growth benefits.

Typically the pH in the upper gastrointestinal tract of healthy males and females is often low enough to counteract the neutral pH of creatine monohydrate (ie. pH ~7).  However, research shows after the administration of a solid and liquid meal gastric pH reached a peak of 6.7 pH and then declined gradually back to a fasted state after approximately two hours.  This data suggests supplementing with creatine monohydrate in a fed state could affect absorption.  Creatine HCL is a suitable alternative due to its lower pH value.

Trimethylglycine (TMG), typically referred to as betaine anhydrous, is derived from the beetroot and is an active metabolite of choline in the body. It serves a vital role in methylation in the body, particularly of the amino acid homocysteine to form methionine. Methionine contributes to the synthesis of creatine in the body.  Therefore supplementing with betaine will lead to an increase in muscle strength through its ability to increase creatine production in the body.  It also been found to increase protein synthesis.

One study, involving 12 trained men (age 19.7 ± 1.23 years), examined the effect of betaine supplementation on selected circulating hormonal measures and Akt muscle signaling proteins after an acute exercise session over the course of 2 weeks.  Betaine vs. placebo supplementation enhanced both the anabolic endocrine profile and the corresponding anabolic signaling environment, suggesting increased protein synthesis.

Twenty-three experienced strength trained men,  matched for training experience (4.8 ± 2.3 years) and body fat percentage (BF%: 16.9 ± 8.0%) were randomly assigned to either a placebo (PL; n = 12) or betaine group (BET; n = 11; 2.5 g/day), and completed a 6 week periodized training program to assess the effect of long term betaine supplementation on body composition, performance, and homocysteine thiolactone (HCTL).  Arm CSA increased significantly (p < .05) in BET but not PL. No differences existed between group and time for changes in thigh CSA. Back squat training volume increased significantly (p < .05) for both groups throughout training. Bench press training volume was significantly (p < .05) improved for BET compared to PL at microcycles one and three. Body composition (BF%, FM, LBM) improved significantly (p < .05) in BET but not PL.  A significant interaction (p < .05) existed for HCTL, with increases from BL to week 2 in PL, but not BET. Additionally, HCTL remained elevated at week 4 in PL, but not BET.  The study concluded that six-weeks of betaine supplementation improved body composition, arm size, bench press work capacity, attenuated the rise in urinary HCTL, and tended to improve power.

L-carnitine is synthesized from the amino acids lysine and methionine and performs

essential functions in human metabolism.  L-carnitine supplementation has been found to have a beneficial effect on markers of postexercise metabolic stress and muscle damage.

A study involving eight healthy males examined the required dose of l-carnitine to elicit a reduction in postexercise metabolic stress and muscle damage.  The study participants were given doses of 0g, 1g, and 2g of LCLT for 3 weeks and then performed a bout of resistance exercise (5 sets of 15-20 repetition maximum with a 2-min rest between sets) with associated blood draws. Serum carnitine concentrations increased (p < or = 0.05) following the 1g and 2g doses, with the 2-g dose providing the highest carnitine concentrations. The 1g and 2g doses reduced postexercise serum hypoxanthine, serum xanthine oxidase, serum myoglobin, and perceived muscle soreness. In conclusion, both the 1g and 2g doses were effective in mediating various markers of metabolic stress and of muscle soreness. Use of LCLT appears to attenuate metabolic stress and the hypoxic chain of events leading to muscle damage after exercise.

18 healthy and active subjects (9 female and 9 male) participated in a double-blind, placebo, balanced treatment presentation and crossover design where they ingested 2 g/d of l-carnitine for 24 days to examine markers of performance and recovery from physical exertion.  Two grams of l-carnitine supplementation had positive effects and significantly (P < or = .05) attenuated biochemical markers of purine metabolism, free radical formation, muscle tissue disruption, and muscle soreness after physical exertion

In a double blind, placebo-controlled study, 18 generally healthy adults were given either a 150 mg dose of elevATP® or 50 mg of an encapsulated silica oxide (placebo).  The researchers measured an acute increase in blood levels of ATP by 64% (P=0.02) when compared to the placebo group.  In addition, the ROS and lactate level in the supplemented group were unchanged.

In a randomized, placebo-controlled, crossover design, seven men with at least two years of resistance training experience ingested 600mg A-GPC (as AlphaSize®) or a placebo 90-minutes prior to completing 6 sets × 10 repetitions of Smith Machine squats at 70% of their predetermined 1-repetition maximum.   Measurements were taken 30 minutes post-exercise and then the subjects performed three sets of bench press throws at 50% of their predetermined 1-repetition maximum to assess peak force, peak power, and rate of force development. All trials were performed after an overnight fast, a 48-hour abstention from intense exercise, and during the same time of day to minimize diurnal variation.  Compared to baseline (pre) values, peak GH increased 44-fold during A-GPC (from 0.19 ± 0.06 to 8.4 ± 2.1 ng/mL) vs. 2.6-fold during placebo (from 1.9 ± 0.8 to 5.0 ± 4.8 ng/mL, P < 0.03) (Figure 3). Peak bench press force was 14% greater in A-GPC (933 ± 89 N) vs. placebo (818 ± 77 N, P < 0.02).

L-glutamine is an amino acid that comprises more than 60% of skeletal tissue.  Supplementation with l-glutamine is believed to reduce recovery time and aid in protein synthesis.  Glutamine has been shown to inhibit protein breakdown in rat skeletal muscle.  However, a human study conducted seems to refute that assertion.  But let’s not write off glutamine just yet because recent research has shown that supplementation of l-glutamine with the amino acid l-alanine can alleviate this issue hence the inclusion of both in TR1UMPH to help your body recover.
